Wall Drug, near Rapid City, SD. Just a giant tourist stop, but the signs for it start 500 miles out in eastern Minnesota.

The Titanic museum and model a guy in Ohio built for himself. It was extremely accurate.

The theatre where John Dillinger was shot, Chicago (OK, not so much "roadside" but just my cup of tea)
